ABSTRACT

Objective To investigate the feasibility of 18G trocar for central venous catheterization in adults. Methods Retrospective analyzed the clinical data of 60 patients with central venous catheterization under local anesthesia. These patients were randomly divided into the control group and the observation group.Selected the internal jugular vein as the site of the puncture catheter. The control group was punc-tured by conventional puncture needle while the observation group was punctured by the 18G trocar. Results There was no statistical differ-ence between the two groups in the success rate of final puncture and catheterization(P>0.05).Compared with the control group, the suc-cess rate of guidewire insert was higher, the puncture operation time was shorter, the pain score during puncture operation was lower, and post-operative patient satisfaction was higher in the observation group. The differences of the two groups were statistically significant(P<0.05). Conclusion 18G trocar can perform central venous catheterization successfully. This method has great advantages for awake patients, and it also worked in some cases with difficult wire insertion.

ABSTRACT

BACKGROUNDNon-small cell lung cancer (NSCLC) is a prolific and high-mortality disease with few effective treatments. Although the detection and surgical techniques for NSCLC continue to advance, the survival rate for the patients with NSCLC remains poor. Enhanced predictive biomarkers such as microRNAs (miRNAs) are needed at the time of diagnosis to better tailor therapies for patients. This study focused on the expression of miR-1280 in NSCLC tissues and distal normal tissues in order to explore the association between miR-1280 expression and NSCLC.

METHODSA total of 72 newly diagnosed primary NSCLC patients were enrolled in this study. Quantitative real-time polymerase chain reaction (PCR) was performed to identify the expression level of miR-1280 in the NSCLC tissues and distal normal tissues of these patients.

RESULTSThe miR-1280 expression was significantly higher in the NSCLC tissues (0.084 ± 0.099) than distal normal tissues (0.014 ± 0.015, P = 0.009). In 54 patients (75%), the miR-1280 expression in the NSCLC tissues was upregulated (2-ΔΔct > 2), and no case showed a downregulation of miR-1280 expression.

CONCLUSIONSThe expression level of miR-1280 could be regarded as a biomarker for NSCLC.

ABSTRACT

OBJECTIVETo study the impacts of exposure to electromagnetic radiation (EMR) on liver function in rats.

METHODSTwenty adult male Sprague-Dawley rats were randomly divided into normal group and radiated group. The rats in normal group were not radiated, those in radiated group were exposed to EMR 4 h/ d for 18 consecutive days. Rats were sacrificed immediately after the end of the experiment. The serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST), and those of malondialdehyde (MDA) and glutathione (GSH) in liver tissue were evaluated by colorimetric method. The liver histopathological changes were observed by hematoxylin and eosin staining and the protein expression of bax and bcl- 2 in liver tissue were detected by immunohistochemical method. Terminal-deoxynucleotidyl transferase mediated nick and labelling (TUNEL) method was used for analysis of apoptosis in liver.

RESULTSCompared with the normal rats, the serum levels of ALT and AST in the radiated group had no obvious changes (P>0.05), while the contents of MDA increased (P < 0.01) and those of GSH decreased (P < 0.01) in liver tissues. The histopathology examination showed diffuse hepatocyte swelling and vacuolation, small pieces and focal necrosis. The immunohistochemical results displayed that the expression of the bax protein was higher and that of bcl-2 protein was lower in radiated group. The hepatocyte apoptosis rates in radiated group was higher than that in normal group (all P < 0.01).

CONCLUSIONThe exposure to 900 MHz mobile phone 4 h/d for 18 days could induce the liver histological changes, which may be partly due to the apoptosis and oxidative stress induced in liver tissue by electromagnetic radiation.

ABSTRACT

OBJECTIVETo analyze the clinical features and gene mutation of a patient with dyskeratosis congenita, who was admitted in our hospital for thrombocytopenia.

METHODThe clinical and laboratory data of a 4 years and 10 months old boy were summarized. DKC1 gene was analyzed using PCR amplification and DNA sequencing.

RESULTThe age of onset of the boy was 1 year. He presented with abnormal cutaneous pigmentation, nail dystrophy and mucosal leukoplakia accompanied by multi-system abnormalities. DKC1 (1058C-T, A353V) was detected in the patient.

CONCLUSIONThe patient presented with classical features of dyskeratosis congenita and DKC1 (1058C-T, A353V) did exist in this patient. X-linked recessive dyskeratosis congenita was confirmed.

ABSTRACT

BACKGROUNDTo understand the significance of detection of serum sialic acid (SA) and Epstein-Barr virus VCA-IgA (EBV-CA-IgA) in diagnosis and monitoring radiotherapy effectiveness of nasopharyngeal carcinoma (NPC) patients.

METHODSSerum SA and EBV-CA-IgA were detected in 65 cases with NPC before radiotherapy and one months after radiotherapy and 21 cases one year after radiotherapy for NPC with local recurrence and/or distant metastasis. Healthy persons and patients with benign lesions of head and neck were also enrolled as control group.

RESULTSSA and EBV-CA-IgA of NPC patients before radiotherapy were significantly higher than those in control group (P<0.01). The sensitivity of combination of SA and EBV-CA-IgA (96.9%) was higher than those determined alone (P<0.05). The SA level of NPC patients after radiotherapy and without recurrence after radiotherapy was reduced significantly compared to the NPC patients before radiotherapy (P<0.01). The SA level of NPC patients with recurrence was significantly higher than that in NPC patients without recurrence (P<0.01), whereas the positive rate of EBV-CA-IgA changed little.

CONCLUSIONDynamic detection of serum SA may be a valuable technique for diagnosis and monitoring radiotherapy effectiveness in NPC patients. The combined determination of the two indexes can raise the positive rate of patients with NPC.

ABSTRACT

AIMTo study the effect of taurine (Tau) on rabbit cardiomyocyte apoptosis during ischemia/reperfusion (I/R) injury.

METHODSRabbit heart I/R injury was induced by occluding the left anterior descending coronary artery for 45 min and reperfusion for 180 min. taurine (200 mg/kg) was intravenously injected 5 min before heart ischemia. Cardiomyocyte apoptosis was measured by using terminal deoxynucleotidyl transferase--mediated dUTP nick end labeling method (TUNEL), flow cytometry (FCM) and DNA agarose gel electrophoresis.

RESULTSDNA ladder pattern of DNA in myocardium was revealed by agarose gel electrophoresis in I/R group while was not found in Tau + I/R group. Apoptotic cardiomyocytes were sparse within ischemic myocardium at risk in Tau + I/ R group as compared with that in I/R group (TUNEL stain). Apoptosis rate in ischemic myocardium from I/R and Tau + I/R groups detected by flow cytometry was 17.66% +/- 1.54% and 4.86% +/- 1.23%, respectively. Fas and Bax protein expressions in ischemic myocardium of I/R group were higher than that in nonischemic myocardium group (P < 0.01), Bcl-2/Bax ratio in I/R group was lower than that in nonischemic myocardium (P < 0.01); while in Tau + I/R group, Fas and Bax protein expressions were lower than that in I/R group (P < 0.01), the Bcl-2/Bax ratio was higher than that in I/R group (P < 0.01).

CONCLUSIONTaurine reduced apoptosis of myocytes in I/R rabbit heart; its mechanism may involve Fas, Bax and Bcl-2 proteins expression.
